Pepsin

Pepsin Powder Manufacturer | High Activity Digestive Enzyme Headline: High Activity Pepsin Manufacturer | Digestive Enzyme API (CAS: 9001-75-6) Sub-headline: 1:3000 / 1:10000 Activity | USP/EP/BP Standards | Porcine Stomach Mucosa Origin Unique Selling Proposition (USP): • High Potency: Extracted from healthy porcine gastric mucosa with activity levels up to 1:10000 NF. • Digestive Power: Breaks down proteins into peptides efficiently in acidic environments (pH 1.5 - 2.5). • Versatile Application: Critical for pharmaceutical digestive aids, food processing (meat tenderizing), and leather bating.

Introduction

P&K

Pepsin (CAS: 9001-75-6) is a potent proteolytic enzyme naturally secreted by the gastric glands. It is one of the main digestive enzymes in the digestive system of humans and many other animals, where it helps digest the proteins in food.

Our Pepsin is isolated from the gastric mucosa of pigs (Porcine) under strict sanitary conditions. It functions optimally in acidic environments, making it an essential ingredient for hydrochloric acid/pepsin supplements designed to treat hypochlorhydria (low stomach acid) and improve protein absorption.

Functions

1.Protein Digestion (Pharmaceutical)

• Mechanism: Endopeptidase activity that cleaves peptide bonds, preferentially aromatic amino acids (phenylalanine, tryptophan, tyrosine).

• Action: Used in digestive syrups and tablets to aid patients with gastric enzyme deficiency, relieving bloating and indigestion.

2.Food Processing

• Mechanism: Controlled hydrolysis of muscle fibers.

• Action: Acts as a meat tenderizer and is used in the pre-digestion of cereals and vegetable proteins to hydrolyze active proteins.

3.Laboratory & Industrial Use

• Mechanism: Specific cleavage of antibodies.

• Action: Used in biology labs to prepare F(ab')2 fragments from IgG antibodies. Also used in leather industry for bating (softening) hides.

Applications

1.Digestive Aid Pharmaceuticals

• Application: Tablets, Capsules, and Syrups (often combined with Betaine HCl).

• Why it works: Replicates the natural gastric environment to break down tough dietary proteins.

2.Food & Beverage

• Application: Curdling agent for cheese (rennet substitute) and hydrolyzing agent for soy protein.

• Why it works: Reduces allergenicity of proteins and improves solubility.

3.Biological Research

• Application: Cell culture and antibody preparation.

• Why it works: High specificity and purity allow for precise cleavage without destroying the target molecule.

Flow Chart

1.Collection: Fresh porcine stomachs harvested from inspected slaughterhouses.

2.Extraction: Acid extraction of the mucosa lining to release pepsinogen.

3.Activation: Conversion of inactive pepsinogen to active pepsin at low pH.

4.Filtration: Removal of insoluble tissues and lipids.

5.Concentration: Vacuum evaporation or ultrafiltration.

6.Purification: Salt precipitation or chromatography to achieve specific activity (e.g., 1:3000).

7.Drying: Spray drying or lyophilization.

8.Standardization: Blending with lactose or other carriers to exact activity specs.

9.Packaging: 25kg Drums.

Method of Analysis of Lactoferrin

Test Method: USP / NF Activity Assay

• Procedure: Determines the digestive power of the enzyme by digesting a standard substrate (coagulated egg albumin or hemoglobin) under controlled acidic conditions.

• Calculation: The activity is expressed as the ratio of the weight of the enzyme to the weight of the substrate digested (e.g., 1:3000 means 1g of pepsin digests 3000g of egg albumin).

Reference Chromatogram of Lactoferrin Reference Substance

(Note: Enzymatic activity is the primary release criteria, not HPLC purity)

SDS-PAGE electrophoresis is used to verify the molecular weight (approx. 35 kDa) and purity of the protein bands, ensuring no contamination with other gastric enzymes like chymosin.

Stability and Safety

Stability Studies

• Thermostability: Unstable above 60°C. Best stored cool.

• pH Stability: Active at pH 2-4. Inactive > pH 6. Irreversibly denatured at pH > 8.

• Shelf Life: 24 Months when stored at 2-8°C or cool room temperature in tight containers.

Safety & Handling (MSDS Summary)

• Classification: Sensitizing substance (Enzyme).

• Handling: Avoid inhaling dust. Enzymatic dust can cause asthma or allergic reactions. Wear N95 mask and gloves.

• Storage: Store in a dry place. Moisture promotes degradation.

FAQ

What does 1:3000 mean?

It refers to the digestive power: 1 gram of this Pepsin can digest 3,000 grams of coagulated egg albumin under standard test conditions. We also supply 1:10000 and custom activities.

Is your Pepsin Halal/Kosher?

Standard Pepsin is extracted from Pig (Porcine) stomachs, so it is NOT Halal or Kosher. We do offer microbial or fungal proteases as Halal/Kosher substitutes if needed.

What is the source?

It is strictly sourced from healthy porcine gastric mucosa from veterinary-inspected slaughterhouses, ensuring TSE/BSE safety.
